摘要
为研究冰糖、葡萄糖对药物体内药动学的影响 ,分析和探讨一些含冰糖、葡萄糖成份的剂型中冰糖、葡萄糖可能起的作用 ,本研究采用荧光光度法测定家兔单独口服 (灌胃 )四环素和合并冰糖、葡萄糖口服 (灌胃 )以后的体内血药浓度及药动学参数的变化。结果显示 ,合并使用冰糖、葡萄糖不仅可使家兔体内血清四环素的 Cm ax比单独口服四环素提高 6倍和 2 .7倍 ,AUCo- 8提高了 4.1倍和 2 .1倍 ,而 Ka、K变化不大。提示冰糖、葡萄糖可显著影响四环素在体内跨膜转运过程 ,可能促进四环素吸收并延缓四环素的排泄。
To investigate the feasible effects of crystal sugar (Bing Tang),a traditional Chinese medicine commonly appeared in compound prescription of Chinese medicine, and glucose on the pharmacokinetics of other drugs and,futher,to explore or reveal what rols or action may crystal sugar and glucose play in some Chinese compound medicine.The present study was carried out to demonstrate the effects of crystal sugar or glucose on the plasma concentrations of tetracycline after single oral dosing.Tetracycline hydrochloride (0.208mg/kg bwt)was administered to the rabbits with or without coadministration of crystal sugar or glucose (3.57mg/kg bwt)by garage (tube feeding).Tetracycline plasma concentrations were determined by fluorophotometric method and the pharmacokinetic parameters were caculated for the comparisons.The results demonstrated that coadministration of crystal sugar or glucose could increase plasma peak concentration of tetracycline by 6.0 and 2.7 folds,the AUC 0-8 by 4.1 and 2.1 times to that of the control, respectively. Although Ka and K value was slightly decreased and increased,respectively,the Tmax was unchanged,this indicates that crystal sugar and glucose enhances the absorption of tetracycline from i.g.tract,and hence increases its bioavailbility.
