摘要
目的 :为了获得血红素氧化酶 - 1基因的 c DNA片段表达载体。方法 :从正常鼠脾细胞提总 RNA进行 RT- PCR、分子克隆 ,并进行自动测序。结果 :分离并鉴定了该 c DNA重组子 ,成功的将血红素氧化酶 - 1基因亚克隆到 pc DNA3.1真核表达载体上。结论 :建立了一个编码血红素氧化酶 - 1基因 c DNA重组子 (76 2 bp)的真核表达载体。
Objective:To obtain expressing vector of the cDNA sequence encoding heme oxygenase 1 gene.Methods:Total RNA from spleen of rat was extracted,RT PCR and cloning were carried out.The sequence was determined with automated DNA sequencing.Results:The cDNA recombinant was sequenced and characterized.These cDNA fragments were ligated into BamH Ⅰ and Hind Ⅲ sites of pcDNA3.1 vector.Conclusion:Expressing vector of the cDNA sequence encoding heme oxygenase 1 gene(762bp) has been determined.
