摘要
目的:观察小蘖碱对氧化型低密度脂蛋白(oxLDL)诱导的大鼠胸主动脉平滑肌细胞(VSMCs)泡沫化及凝集素样氧化型低密度脂蛋白受体1(LOX-1)、核转录因子P65(NF-κB P65)表达的影响。方法:将一部分体外培养的VSMCs分为空白对照组,泡沫化模型组(简称模型组),50μM小蘖碱对照组和50μM小蘖碱干预组,油红O染色后,观察各组VSMCs的泡沫化程度。另一部分体外培养的VSMCs同上分为空白对照组、模型组以及25μM小蘖碱干预组和50μM小蘖碱干预组,预刺激后Western-Blotting检测各组VSMCs的LOX-1蛋白和NF-κB P65蛋白表达水平。结果:空白对照组和小蘖碱组VSMCs未泡沫化,LOX-1和NF-κB P65微量表达。模型组VSMCs中的脂滴聚集成团,形成泡沫化,LOX-1及NF-κB P65的表达较对照组明显增加(P<0.01),小蘖碱干预组泡沫化明显减轻,两小蘖碱干预组VSMCs LOX-1及NF-κB P65蛋白表达较模型组显著降低(P<0.01),高效量组作用更强(P<0.05,P<0.01)。结论:小檗碱可减轻平滑肌细胞源性泡沫细胞化程度,其作用机制可能与抑制NF-κB和LOX-1蛋白表达有关。
Objective:To investigate the effect of berberine on oxLDL induced foam cell formation and lectin-like oxidized low density lipoprotein receptor1(LOX-1),NF-κB expression in vascular smooth muscle Cells(VSMCs).Method:VSMC were cultured and classified into four groups:control group,foam cell model group(treated with oxLDL for48hto establish the foam cell models),or 25uM berberine group and 50uM berberine group(different dose of berberine was pretreated for 2hours).LOX-1protein expression was detected by western-blot after treated with oxLDL for 48h.Western-blot was applied to detect nuclear expression of NF-κB P65protein after treated with oxLDL for 12h.Results:Compared with the foam cell models,oxLDL agregation and lipid droplets of foam cells in berberine treatment decreased significantly.The LOX-1protein expression decreased at a low ebb after berberine treatment(P〈0.05).On the other hand,with the increasing concentration of berberine treatment,the LOX-1protein expression decreased progressively(P〈0.01).With two dosages of berberine,significantly difference was shown between berberine groups and oxLDL group for nuclear NF-κB P65protein expression(P〈0.01).Conclusion:Berberine inhibits LOX-1and nuclear NF-κB P65protein expression in a dose-dependent manner.This may be part of the mechanism of its anti-atherosclerotic effect.
