摘要
目的探讨阿托伐他汀对实验兔动脉粥样硬化模型血脂、血管内皮生长因子(VEGF)水平及动脉粥样斑块内VEGF蛋白水平的影响。方法 20只新西兰大白兔,随机分为对照组5只,高脂组15只;高脂组喂养4 w,用球囊损伤腹主动脉后,随机分为模型组、低剂量他汀组、高剂量他汀组,每组各5只,共饲养12 w。ELISA检测各组血清TG、TC、LDL-C、VEGF;12 w末检测动脉粥样斑块内VEGF蛋白定量及VEGF mRNA水平。结果与对照组相比,模型组、他汀组6 w和12 w血清LDL-C、TG、TC和VEGF均显著增高(P<0.01);与模型组比较,他汀组血脂和血清VEGF明显降低(P<0.05);与低剂量他汀组比较,高剂量他汀组血脂和血清VEGF明显降低(P<0.05);与模型组比较,他汀组动脉粥样硬化斑块中VEGF蛋白水平下调,但高剂量他汀组显著降低动脉粥样硬化斑块中VEGF的表达(P<0.05)。结论兔动脉粥样硬化模型血清VEGF水平及动脉粥样硬化斑块内VEGF高表达,阿托伐他汀可下调AS斑块内VEGF的表达水平。
Objective To study the effects of atorvastatin on serum lipid and vascular endothelial growth factor（ VEGF） and its effects on VEGF in atherosclerotic plaque in rabbit model. Methods 20 New Zealand rabbits were randomly divided into 2 groups：control group（ n = 5） and hyper-cholesterol group（ n = 15）. The rabbits in hyper-cholesterol group were fed with hyper-cholesterol diet for 4 weeks and then catheter-induced abdominal aorta injury was made to them; then the 15 rabbits were randomly subdivided into 3groups： model group（ n = 5）; low-dose atorvastatin group（ n = 5） and high-dose atorvastatin group（ n = 5）. All rabbits were kept feeding for 12 weeks. The enzyme-linked immunosorbent assay was applied to measure the serum TC,TG,LDL-C and VEGF of animals at the end of the 12th week; VEGF protein and mRNA in atherosclerotic plaque were detected. Results The serum LDL-C,TG,TC and VEGF in model group and atorvastatin groups at the 6th and 12th week were higher than those in control group（ P〈0. 01）; the serum lipid and VEGF in high-dose atorvastatin group decreased more than those in low-dose group（ P〈0. 05）; the level of VEGF protein in atherosclerotic plaque of the rabbits in atorvasatin groups dropped down more than that of the rabbits in model group and the decrease in high-dose group was more obvious（ P〈0. 05）. Conclusions The expression level of VEGF is high in serum and atherosclerotic plaque of rabbit＇ model and atorvastatin can down-regulate the expression level of VEGF in atherosclerotic plaque.
