测定异烟肼血药浓度RP-HPLC的建立及其应用 被引量:2

摘要 目的为了探讨治疗结核性脑膜炎时异烟肼的剂量选择,建立一种快速、灵敏的测定血浆中异烟肼药物浓度的反相高效液相色谱法(RP—HPLC)。方法替硝唑作内标,经与香草醛衍生化反应后进样。用Symme.tryC18(4.6mm×250mm,5um)反相色谱柱进行分离,以甲醇-0.02mol·L-1磷酸二氢钾溶液(体积比为39:61)为流动相,流速1.0mL·min-1,柱温为30℃,检测波长为340nm,20ul进样。结果异烟肼血药浓度在0.1~16ug·mL-1(r=0.9999)范围内线性关系良好;日内、日间RSD均〈15%,提取回收率60.63—66.95%。结论本方法简便、准确、灵敏,满足异烟肼血药浓度测定,可应用于临床结核性脑膜炎患者血药浓度的检测及药代动力学分析。 Objective To discuss the dose strategies of isoniazid in the treatment of tuberculous meningitis to establish a method of RP - HPLC on the determination of plasma concentrations of isoniazid. Methods With tinidazole as an internal standard,the isoniazid and vanillin were injected after their derivatization reaction. Separated by reversed phase chromatographic column of Symmetry - C18 (4.6 mm × 250 mm,5 um), the mobile phase was consisting of 0.02 mol · L-1 potassium dihydrogen phosphate buffer and methanol ( the volume ratio of them was 61 - 39 ). The total mobile phase rate was 1.0 mL · min -1 , column at 30 ℃, and the detection wavelength was set at 340 nm, and injected volume was 20 uL. Results The plasma concentrations of isoniazid had a good linear relationship within the range of 0.1 - 16.0 ug · mL- 1 ( r = 0. 999 9). The RSD of intra - day and inter - day were both below 15%, and the coefficient of recovery was 68.60% - 75.00%. Conclusion The method of this study is simple, accurate and sensitive,and it can be used to detect isoniazid plasma concentrations in clinical patients with tuberculous meningitis and to investigate the pharmacokinetics of isoniazid.
