摘要
目的:建立高效液相色谱法同时测定人血浆中异烟肼、乙酰异烟肼和吡嗪酰胺的浓度。方法:色谱柱为Inertsil ODS-SP(4.6 mm×150 mm,5μm),以甲醇∶0.05 mol·L-1磷酸二氢钾(三乙胺调pH 5.6)为流动相,柱温:40℃,流速:1.0 mL·min-1,进样体积:20μL,检测波长:268nm。结果:3组分在线性范围内与峰面积线性关系良好(r2〉0.9940),日内、日间精密度RSD〈10%,平均回收率在80%~120%。结论:本方法专属性好,样品处理简单,符合生物样品分析要求。
Objective:To determine the concentrations of isoniazid,acetylisoniazid and pyrazinamide in human plasma by RP-HPLC.Methods:The HPLC method was carried out on an Inertsil ODS-SP(4.6× 150 mm,5 μm) column with 0.05 mol·L-1KH2PO4(adjusting PH to 6.0 with triethylamine)-methanol as mobile phase.The column temperature was 40 ℃,the flow rate was 1.0 mL·min-1and the detection wavelength was 268 nm.Results:All calibration curves had good linearity(r2>0.99) within their tested ranges.The intra-and inter-day precision was within 10%,the extraction recovery was between 80% and 120%.Conclusion:The method is sensitive,simple and rapid.It is suitable for analyzing the drugs in biological samples.
