摘要
目的观察化合物48/80(Compound 48/80,C48/80)对正常和TRPV1敲除小鼠诱发的瘙痒反应及电针不同穴位对其的影响,明确针灸止痒效应及与TRPV1的关系。方法选用野生型C57BL/6小鼠(WT小鼠)和TRPV1敲除小鼠(KO小鼠)各30只,分为模型组、曲池组、血海组,连续两日分别于其颈部中央皮下注射C48/80,曲池组、血海组于第二日注射前给予电针治疗30min,观察电针干预前后瘙痒行为即搔痒次数/30min的变化。结果 C48/80能诱发WT小鼠和KO小鼠对注射处显著的搔抓反应,模型组WT小鼠第一日的搔痒次数为(180.69±79.00)次/30min,KO小鼠第一日的搔痒次数为(124.87±31.52)次/30min,二者差异具有统计学意义(P<0.05)。电针曲池、血海均能有效抑制此种搔抓反应(P<0.01,P<0.05),曲池组WT小鼠的电针前后变化率为49.66%,KO小鼠为35.66%,二者差异具有统计学意义(P<0.01);血海组WT小鼠为52.72%,KO小鼠为34.91%,二者差异具有统计学意义(P<0.01)。而对WT与KO小鼠而言,穴位间止痒效应的差异无统计学意义(P>0.05)。结论 TRPV1参与C48/80诱发的瘙痒反应的产生,电针能明显抑制化合物48/80诱发的小鼠皮肤瘙痒反应,且TRPV1参与针刺效应的产生。
Objective To observe the effect of electroacupuncture (EA) at Quchi (LI11 ) or Xuehai (SP10) on experimental cutaneous pruritus induced by compound 48/80 in mice. Methods 30 BALB/c mice were randomly divided into model group, Quchi group and Xuehai group. Compound 48/80 was injected subcutaueously into the rostral part of the back in two consecutive days. Quchi group and Xuehai group were given electro - acupuncture treatment for 30min before injection of the second day. The number of scratches was counted for 40 min after injection of compound 48/80. Itching behavior was observed before and after the intervention of the electro - acupuncture. Results The scratching reaction can be significantly induced in mice after the injection of compound 48/80. Acupuncture at Quchi(LI11 ) and Xuehai (SP10) caused a potent depressant effect on compound 48/80 - induced scratching behavior. However there was no significant difference between Quchi ( LI11 ) group and Xuehai group. Con- clusion EA can significantly inhibit skin itching reaction in mice induced by compound 48/80, and may be one of the effective methods for treatment of pruritus. The selection of acupoint is the important factor affecting antipruritic effect of EA.
