摘要
目的 探讨瘢痕疙瘩周边部和中央部细胞生物学差异以解释瘢痕疙瘩呈浸润性生长的机理。方法 取手术切除的瘢痕疙瘩组织各 6例为标本 ,通过细胞培养 6~ 10代后 ,应用流式细胞仪、粘附式细胞仪检测瘢痕疙瘩不同部位成纤维细胞P53 蛋白的表达和细胞周期的分布。结果 瘢痕疙瘩周边部成纤维细胞大量分布于增殖期 (G2 、S、M期 ) ,且P53 蛋白表达较低 ;而瘢痕疙瘩中央部成纤维细胞P53 蛋白呈强表达 ,且主要分布在静止期 (G1、G0 期 )。结论 瘢痕疙瘩不同部位成纤维细胞细胞周期的分布及P53 蛋白表达的差异可能是导致其不同生长特性的细胞生物学机理之一 ,也可能是导致瘢痕疙瘩呈浸润性生长的机理之一。
Objective To explore the concrete machanisms accounting for the different growth characteristics in peripheral areas and central areas of keloid. Methods Six samples of keloids were collected and only 6~10 passages of fibroblasts through cell culture were selected for experiment. The distributions of cell cycle and expression of protein P 53 were analyzed with the help of flow cytometer and adherent cell analysis and sorting interactive laser coytometer(ACAS 570). Results A large percent of fibroblasts derived from peripherial areas of keloids were distributed in G 2、 S and M stages with low levels of P 53 protein. On the other hand, fibroblasts derived from central areas of the keloids distributed mostly in G 0 and G 1 stages with a high expression of P 53 protein. Conclusion The difference in distributions types of cell cycle and expression of protein P 53 may account for the different growth characteristics in peripheral vs. Central areas and may be possiblly one of machanisms accounting for the aggressive growth of keloid.
