雷公藤配伍甘草治疗类风湿关节炎减毒增效作用研究 被引量:10

Toxicity Reducing and Efficacy Enhancing Research on Rheumatoid Arthritis Effect of Tripterygium Compatible with Licorice

摘要 目的:对雷公藤与甘草配伍前后的药效和毒性进行比较研究,初步探讨传统中药配伍减毒增效理论的科学内涵。方法:以昆明种小鼠和Wistar大鼠为试验动物,研究雷公藤配伍甘草前后ig给药对受试动物的镇痛、抗炎作用及毒性的影响。采用热板法和醋酸致小鼠扭体反应进行镇痛实验;采用角叉菜胶致大鼠足跖肿胀及DNCB引起的小鼠迟发性超敏反应进行抗炎药效学实验。结果:雷公藤与甘草配伍前后均能明显延迟小鼠舔后足时间,并能减少醋酸引起的小鼠扭体反应,抑制率均在24%以上;对角叉菜胶引起的足跖肿胀,1~4h的抑制率在21%以上;对DNCB引起的小鼠迟发性超敏反应有明显的抑制作用,抑制率为47.9%,且与甘草配伍后雷公藤的药理作用有显著提高。结论:甘草可以明显增强雷公藤的抗炎、镇痛作用,提示中药的配伍减毒增效作用可通过降低毒性药物用量,并配伍增效药物而实现。 Objective:Compare the pharmacokinetics and toxic between Tripterygium and Licorice, to investigate the traditional Chinese medicine theory on toxicity reducing and efficacy enhancing. Methods : The pharmacodynamic effects were observed in 2 an- algesic tests and 2 inflammatory models in experimental animals,including: hot plate test in mice, writhing reaction induced by acetic acid in mice, paw swelling caused by carrageenan in rats and auricular edema induced by DNCB in mice. Results: Tripterygium could delay the latency of licking of the hind paw in hot plate test. And it could reduce the times of writhe response actions, and the inhibition rates were more than 24% Tripterygium could significantly inhibit the paw swelling caused by carrageenan in rats, and the inhibition rates were more than 21 % from 1 to 4 hour. And then it could significantly inhibit the Delayed hypersensitivity induced by DNCB, and the inhibition rates were 47.9 %. Furthermore the effect above could be additional increased, when tripterygium com- patibitied with Licorice. Conclusion:Licorice can significantly enhance anti-inflammatory, analgesic effect of tripterygium, but can not reduce the toxic of tripterygium. The phenomenon shows that the theory on toxicity reducing and efficacy enhancing in Chinese compatibity may achieve via reducing the content of toxic medicine and compatibitying another medicine at the same time.
