百草枯中毒鼠肺组织中TM-EPCR mRNA与IL-1β的表达 被引量:2

摘要 目的探讨IL-1β与TM、EPCR基因在百草枯中毒鼠所致急性肺损伤中的表达.方法 90只SD雄性大鼠分为正常对照组(C组30只)、百草枯中毒组(PQ组60只).PQ组一次性灌胃PQ25 mg/kg染毒,C组予等体积生理盐水灌胃.观察各组大鼠在中毒后6 h、12 h、1 d、3 d、5 d、7 d肺组织病理改变,采用反转录-聚合酶链反应(RT-PCR)方法检测鼠肺组织TM、EPCR mRNA的表达;采用ELISA方法检测血清的IL-1β表达.结果 C组肺组织结构完整,无炎症细胞渗出.PQ组肺组织炎性细胞浸润明显增多,且随时间推移肺损伤加重.C组TM和EPCR mRNA有一定水平表达;与C组相比PQ组鼠肺组织TM和EPCR mRNA的表达增强,差异有统计学意义(P<0.05);PQ组TM和EPCR mRNA的表达在1 d达高峰之后下降,第3天基本恢复到C组水平.PQ组IL-1β与TM表达呈正相关(P<0.01);IL-1β与EPCR表达呈正相关(P<0.01).结论 IL-1β与TM、EPCR基因介导的炎症反应参与百草枯中毒所致肺损伤. Objective To explore the expressions of thrombomodulin (TM) and endothelial protein C receptor (EPCR) mRNA and IL-1β in lung of paraquat poisoning rats. Methods Ninty SD male rats were randomly divided into 2 groups: normal control group (C30) and paraquat poisoning group (PQ60) The PQ group rats were given paraquat by gastric perfusion (25 mg/kg) ; The C group rats were given saline by gastric perfusion. Histological changes of lung tissues under light microscope were observed. And the expression of TM and EPRC mRNA in lung tissues of rats were measured by RT-PCR at different time of 6h, 12h, ld, 3d , 5d and 7d after poisoning. The expression of IL-1 β was determined by ELISA. Results The relative magnitude expression of TM and EPCR mRNA in PQ (6 h), PQ (12 h), PQ (ld) and PQ (3d) subgroups were significantly higher as compared with the C group (P〈0.05) . The content of IL-1 13 in PQ group was higher than that in C group; The pathological scores of lung injury in PQ groups were significantly higher compared with C group. The expression of IL-1 β positively correlated with that of TM (P〈0.01), and positively correlated with that of EPRC (P〈0.01). Gonclosioo The aggravation of lung injury induced by paraquat in rats is related to the expression of IL-1 β and TM, EPCR mRNA in lung tissue.
