摘要
采用包埋-交联法对转谷氨酰胺酶进行固定化反应,以去酰胺改性后的大豆蛋白为底物,利用固定化转谷氨酰胺酶对其进行聚合改性,初步研究了聚合反应的温度,时间,给酶量,pH及振荡仪的转速对改性反应的影响。结果表明,固定化转谷氨酰胺酶的最佳改性条件为:反应温度45℃,反应时间6 h,反应pH为7,给酶量15 U/g蛋白,采用恒温振荡器反应的转速应为125 r/min。海藻酸钠固定化转谷氨酰胺酶可重复利用3次左右。
Transglutaminase was immobilized to raise the utilization rate and utilize transglutaminase time. Proteins were produced to improve features with alginate sodium as immobilized carrier and glutaraldehyde as crosslinker. Immobilized transglutaminase was used as catalyst to polymerize the soybean proteins, the optimum conditions of the catalytic reaction was as followed: the optimum temperature was 45 ~C, reacting 6 h and pH 7, the concentration of the enzyme was 15 U/g protein and the rotating speed was 125 r/min in the constant temperature oscillator. The immobilized transglutaminase could be used 3 times repeatedly.
