摘要
以生长在非控温控光下 9个甘蓝型黄籽油菜 (Brassicanapus)为供体 ,进行花药培养。结果表明 ,甘蓝型黄籽油菜愈伤组织诱导频率受花蕾长度、蔗糖浓度、培养基等因素影响。取材以 2mm~ 3mm的花蕾效果最好 ,培养基则是含6 %~ 10 %蔗糖浓度的B5培养基最好。并发现愈伤组织分化时 ,B5培养基附加 6 - 8A4.0mg/L和NAA0 .2mg/L效果较好。
Nine yellow-seeded varieties of rape( Brassica napus L.) were grown under uncontrolled temperature and light conditions and their anthers were used as explants in in vitro culture.It was shown that callus induction rate was influenced by flower bud size, medium and sucrose concentration in the medium. Flower buds 2mm~3mm in length were most desirable, and B5 supplemented with 6%~10% sucrose appeared to be the optimum medium. Best results in callus differentiation was obtained in B5 medium enriched with 6-BA 4.0 mg/L and NAA 0.2 mg/L.
