甘蓝型油菜(Brassica napus L.)花药培养中若干因素对花粉胚状体诱导和植株再生的影响 被引量:20

摘要 研究表明,甘蓝型油菜(Brassica napus L.)花粉胚状体的诱导频率与培养材料的基因型、培养基蔗糖浓度、培养基附加成分、培养基状态、温度处理等因素有关。不同基因型材料的花粉胚状体诱导频率明显不同;培养基蔗糖浓度以10%为最佳;B_5培养基附加2,4-D 0.1mg/l和NAA0.1mg/l的培养效果最好;液体浅层静止漂浮培养优于固体培养;变温处理可以提高花粉胚状体诱导频率。本文也报道了五种不同的花粉胚状体类型与植株再生的研究。 The major factors affecting the induction frequency of pollen embryoid through anther culture in Brassica napus L. were discussed in this paper. The experimental result showed that anthers from different genotypes had different induction frequencies of pollen embryoids and that 10% concentration of sucrose in medium was the optimum dosage of sucrose. It was observed that B_5 medium added with 2,4-D0 1mg/l plus NAA 0.1 mg/l led to the best result and static suspension culture was superior to agar-solidified culture. Changing temperature treatment could enhance the induction frequency of pollen embryoid. The relationships between five different pollen embryoid types and plant regeneration were also reported here.
