摘要
目的探讨miR-126,miR-130a,miR-222在冠心病患者外周血浆中的表达水平及其临床意义。方法选择冠心病患者79例(45例轻度组、34例重度组),冠状动脉阴性对照组24例,收集两组样本临床资料;运用Real-time PCR法检测样本血浆miR-126、miR-130a、miR-222表达水平;分析其表达差异及与脂蛋白a相关性,评估血浆miR-126、miR-130a、miR-222在冠心病中的表达水平及其临床意义。结果 (1)与对照组相比,冠心病组患者血浆miR-126和miR-222的表达水平降低和升高,分别是对照组的0.08倍和4.23倍,差异有统计学意义(P<0.05);miR-130a表达水平降低,是对照组的0.32倍,但差异无统计学意义(P>0.05)。(2)以改良Gensini评分对冠心病患者进行亚组分析,冠心病轻度组与重度组miR-126的表达水平分别为对照组的0.15倍及0.004倍,miR-222的表达水平分别为对照组的2.77倍及7.38倍,差异均有统计学意义(P<0.05),并且miR-126、miR-222表达水平变化与改良Gensini评分相关。(3)冠心病组中患者Lpa与miR-126、miR-130a、miR-222表达水平均无相关性。结论冠心病患者外周血浆中miR-126、miR-222表达水平较冠状动脉阴性对照组有显著差异,且表达水平变化与冠状动脉狭窄程度密切相关。
Objective To investigate the expression and clinical implication of peripheral plasma miR-126, miR-130a, miR-222 in patients with coronary artery disease(CAD). Methods 79 patients with CAD (CAD group: 45 mild, 34 severe) and 24 controls without CAD were included. General clinical data and related biochemical parameters were collected. Plasma miR-126, miR-130a, miR-222 levels were detected with quantitative real-time polymerase chain reaetion(qRT-PCR) analysis and the expression differences were analyzed. The correlation between miR-126, miR-130a, miR-222 and lipopmtein(a) were analyzed also. Results (1) Compared with control group, the expression level of plasma miR-126 was significantly decreased in CAD group (fold 0.08) and miR-222 was significantly increased in CAD group (fold 4.23)(P〈0.05); miR-130a was decreased (fold 0.32) (P〈0.05). (2) In CAD subgroup of improved Gensini method, the expression level of plasma miR-126 and miR-222 were significantly different in mild and severe group, miR-126 (mild with fold 0.15, severe with fold 0.004) (P〈0.05), miR-222 (mild with fold 2.77, severe with fold 7.23)(P〈0.05). The expression level changes of plasma miR-126 and miR-222 were closely related to improved Gensini scores. (3)There was no correlation between miR-126, miR-130a, miR-222 and lipoprotein(a) in CAD group (P〉0.05). Conclusion Compared with control group, the expression levels of peripheral plasma miR-126 and miR-222 were significant difference in patients with CAD. The expression level changes of plasma miR-126 and miR-222 were closely related to the severity of Coronary artery stenosis.
