食管鳞癌组织中MUC15基因和蛋白质的表达及其与临床病理因素的关系 被引量:4

摘要 目的:探讨MUC15在食管鳞状细胞癌(鳞癌)中的表达及其与鳞癌发生、发展的关系。 方法:采用RT-PCR法及免疫组织化学SP法检测53例正常食管黏膜组织、20例不典型增生组织及53例食管鳞癌组织中MUC15 mRNA的相对含量及蛋白质的阳性表达率,并比较其在不同临床病理因素下的差异。 结果:在正常食管黏膜组织、癌旁不典型增生组织及食管鳞癌组织中MUC15 mRNA相对表达量分别为0.281±0.017、0.322±0.029、0.790±0.038;蛋白质表达阳性率分别为20.7%、35.0%、88.7%,差异均有统计学意义(P〈0.05)。临床分期0~Ⅱ期及Ⅲ~Ⅳ期的食管鳞癌组织中MUC15 mRNA相对表达量分别为0.498±0.029、0.832±0.045;蛋白质表达阳性率分别为70.0%、93.0%,差异均有统计学意义(P〈0.05)。有淋巴结转移组与无淋巴结转移组之间mRNA相对表达量分别为0.829±0.031、0.501±0.042;蛋白质表达阳性率分别为90.2%、75.0%,差异有统计学意义(P〈0.05)。 结论:食管鳞癌组织中MUC15表达量较正常食管组织有明显升高,其异常高表达可能与食管鳞癌的发生、发展有关。 Objective:To investigate the expression of MUC15 in esophageal squamous cell carcinoma (ESCC) and its relationship with the occurrence and development of ESCC. Methods:The mRNA levels and protein expression rates of MUC15 in 53 cases of normal esophageal mucosa tissue, 20 mucosa tissue adjacent to cancer and 53 ESCC tissue samples were determined by RT-PCR and the SP immunohistochemical method, respectively, and the expression difference of MUC15 between different tissues was compared. Results:The relative mRNA levels of MUC15 gene in the normal esophageal mucosa tissue, mucosa adjacent to cancer and ESCC tissue were 0.281±0.017, 0.322±0.029 and 0.790±0.038, respectively, and the protein expression rates 20.7%, 35.0% and 88.7%, respectively, with significant difference between different tissues (P〈0.05). In addition, the relative mRNA levels and protein expression rates of MUC15 in stages 0~Ⅱ and Ⅲ~Ⅳ of ESCC were 0.498±0.029, 0.832±0.045 and 70.0%, 93.0%, respectively, with significant difference between them (P〈0.05). Moreover, there was also significant difference of relative mRNA levels (0.829±0.031 vs 0.501±0.042) and protein expression rates (90.2% vs 75.0%) of MUC15 between ESCC with and without lymph node metastasis (P〈0.05). Conclusion:The expression levels of MUC15 gene and protein in ESCC tissue are significantly higher than that in normal esophageal tissue, indicating that they may be associated with the carcinogenesis and development of ESCC.
