凝血酶-富血小板血浆复合物影响体外培养的人软骨细胞生长的实验研究

摘要 目的探讨凝血酶-富血小板血浆复合物对于体外培养的人软骨细胞生长的影响。方法制备凝血酶-富血小板血浆(PRP)复合物,取第三代软骨细胞,接种于96孔培养板,每组5孔,分为:阴性对照组、阳性对照组、实验组,用cck-8(细胞增殖)法检测各组OD值。结果实验组与阴性对照组、阳性对照组的软骨细胞增殖变化比较,均差异具有统计学意义(P<0.05);且实验组内,凝血酶为60 U时对软骨细胞具有促增殖作用(P<0.05)。结论凝血酶-富血小板血浆复合物能促进体外培养的人软骨细胞的增殖,以60 U/ml为最适浓度,为进一步验证其可靠性,还有待于扩大样本或继续进行相应实验证实。 Objective To investigate the effect of thrombin platelet rich plasma complexes for in vitro cultured human chondrocytes. Methods Preparation of thrombin and platelet rich plasma (PRP) complexes, the third passage chondrocytes, seeded in 96 well culture plates, each of the 5 hole, was divided into: negative control group, positive control group, the experimental group, CCK-8 (cell proliferation) was used to detect the OD value. Results The experimental group and negative proliferation of chondrocyte group, positive control group were compared, there were significant differences, P〈0.05 ; and in the experimental group, thrombin was 60U on chondrocytes proliferation promoting effect (P〈0.05). Conclusion thrombin platelet rich plasma compound can promote the proliferation of ehondroeyte in vitro, but with the concentration of thrombin increased its role in promoting the proliferation of cartilage cells was weakened, low concentrations of thrombin, promote the proliferation of cartilage ceils is better, and the optimum concentration is 60 U/ml.
