摘要
目的 :通过动物体内实验了解治疗剂量阿霉素 (adriamycin ,ADR)注射对心肌细胞胞浆内游离钙浓度的影响 ,并探讨心脏阿霉素中毒的病理生理机制。方法 :实验兔 9只每周注射ADR 1次 ,共 8周。并以健康同龄兔 9只为对照组。测定体重(BW ) ,心室重量 (VW ) ,心室重量 /体重 (VW /BW ) ,颈动脉压 (Bp) ,颈动脉平均压 (MAP) ,左心室收缩压 (LVSP) ,左心室舒张末压 (LVEDP) ,心指数 (CI) ,血浆心钠素 (ANP) ,心肌细胞胞浆内游离钙浓度 (MyoCa2 +)。结果 :用药后实验组BW明显下降 ,但VW /BW高于对照组。同时 ,实验组CI,BP ,MAP ,LVSP均较对照组低 ,而实验组LVEDP、ANP及MyoCa2 +则明显增高。且VW /BW与CI呈负相关 ,MyoCa2 +与CI呈负相关。结论 :治疗剂量阿霉素注射后可使心室肥厚 ,心脏功能下降。心肌细胞内钙超载是阿霉素心功能损伤的重要病理生理机制。
OBJECTIVE:To evaluate influence of treated-dose adriamycin (ADR) on calcium in cardiomyocyte in rabbits.METHODS:Nine rabbits received an injection of adriamycin (2 mg·kg -1 ,i.v.) once a week for eight weeks.Nine rabbits were similarly injected with saline as control groups. The body weight (BW),ventricular weight (VW),blood pressure (Bp),left ventricular systolic pressure (LVSP),left ventricular end-diastole pressure (LVEDP),cardiac index (CI),plasma atrial natriuretic peptide (ANP) and calcium in cadiomyocyte (MyoCa 2+ ) of rabbits after the final injection were examinated.RESULTS:BW,CO,Bp and LVSP were significantly decreased in the ADR treated group compared with the control group. Conversely,VW/BW was higher in ADR treated rabbits.Mean while,CI was negatively correlated with MyoCa 2+ and VW/BW respectively.CONCLUSIONS:Heart function was decreased by treated-dose ADR injecion.Calcium overload in cardiomyocyte is important physiopathologic mechanism in ADR-induced impariment of heart.
