灯盏花素对大鼠岛状皮瓣血液流变学和氧化应激的影响 被引量:6

摘要 目的观察灯盏花素对大鼠岛状皮瓣丙二醛(MDA)、超氧化物歧化酶(SOD)、总抗氧化能力(T-AOC)和血液流变学的影响,探讨灯盏花素对皮瓣缺血再灌注损伤后的保护机制。方法健康清洁雄性SD大鼠108只,平均分为假手术组(SO组)、缺血再灌注组(IR组)和灯盏花素干预组(BR组)。建立大鼠腹部岛状皮瓣缺血再灌注损伤模型。术前30 min分别一次性给予生理盐水(SO组、IR组)和灯盏花素注射液(BR组)腹腔注射。术后检测各组大鼠血液中MDA含量、SOD活性、T-AOC活性及血液流变学指标,并观察皮瓣成活率。结果 IR组、BR组再灌注后4、8及24 h 3个时点MDA含量均高于SO组,SOD、T-AOC活力和血液流变学状态低于SO组(P<0.05);并且BR组在这3个时点的各项指标优于IR组(P<0.05)。皮瓣成活率缺血再灌注组<灯盏花素组<假手术组,差异有统计学意义(P<0.01)。结论灯盏花素能通过改善微循环和提高机体抗氧化的能力,减轻皮瓣缺血再灌注损伤,促进皮瓣成活。 Objective To investigate the effects of breviscapine on malonyldialdehyde ( MDA), superoxide dismutase (SOD) and hemorrheology in rats island skin flap, and explore the protective mechanisms of breviscapine for the skin flap after isehemia reperfusion injury. Methods One hundred and eight male SD rats are randomly classified into sham operation group( SO group) ,ischemia/ reperfusion group( IR group)and breviscapine intervening group( BR group). A rat abdomen island skin flap model was established with ischemia reperfusion. The rats were intraperltoneal injected with breviseapine for the BR group and saline for the SO and IR groups be- fore 30 rain of surgery. The total antioxidant capacity(T-AOC) ,level of MDA and SOD activity were compared among these groups. The survival rate of flaps was also observed by using image analysis. Results The expressions of MDA in the IR and BR group were higher than that in the SO group at 4,8 and 24 hours after reperfusion ,especially in the IR group( P 〈 0.05 ). The activity of SOD ,T-AOC and hemorrheology in the IR and BR group was lower than that in the SO group at 4,8 and 24 hours after reperfusion,especially in the 1R group (P 〈 0. 05 ). The survival rate of flap was IR group 〈 BR group 〈 SO group(P 〈 0. 01 ). Conclusion The intraperitoneal use of breviscapine may promote the survival rate of skin flaps after ischemia reperfusion in rats. The possible protective mechanism may be that breviscapine can expand the microvascular, improve the circulation, reduce the level of oxygen free radical and decrease the flap ischemia-reperfusion injury.
