摘要
对碱裂解法提取质粒DNA的方法进行了一些改进 ,在室温下裂解菌体和变性DNA ;用NH4 Ac和LiCl同步沉淀除去样品中的蛋白质和RNA ,降低了对超螺旋质粒的剪切作用 ,得到高质量的质粒DNA。
Modifications were made on the preparation of plasmid by alkaline lysis. Cells were lysed and DNA were denatured under room temperature, NH 4Ac and LiCl were both used to precipitate proteins and RNA in sample ,destruction of supercoiled DNA were reduced, which yield high quality plasmid DNA.
