摘要
为了提高籼稻的基因枪转化频率 ,以 14种重要的籼稻栽培品种为材料 ,对其胚性愈伤组织的诱导与再生的培养条件、转化细胞的筛选方案以及基因枪轰击参数进行了比较研究 ,建立起了较高优化的基因枪转化系统 ,并获得了一批有价值的转化植株。
In order to improve the frequency of indica rice transformation by biolistic bombardment, the suitable culture conditions for embryonic callus and the optimal selection scheme for resistant calli and seedlings, and the optimum bombardment parameters were investigated by using 14 commercially important indica rice cultivars. The main results showed that the CC medium with 36g/L mannitol is a scheme subculture medium in which the browning of indica rice calli can be mitigated significantly; The concentration of 30~40mg/L hyg or 150~200mg/L G418 or 10~20 mg/L Basta is suitable for selection of resistant calli;The transformation parameters of 100 μg gold powder absorbing 0.2 μg DNA per shot and 900 psi helium pressure and 6 cm bombardment distance and bombarded twice each plate gave the best result; Keeping the target calli on osmotic medinm containing 60g/L mannitol from 12~24h before bombardment to 24~48h after it may increase the efficiencies of transfor Matfon. Furthermore, some transgenic indica rice plants were obtained using this transformatim system optimized by the authors.
