肾移植急性排斥大鼠的血清NGAL和CXCL10表达及意义 被引量:1

摘要 目的:探讨同种异体肾移植大鼠血清的肾损伤因子中性粒细胞明胶酶相关脂质运载蛋白(neutrophil gelatinase-associated lipocalin,NGAL)和趋化因子CXC基序配体10(C-X-C motif chemokine 10,CXCL10)表达与急性排斥反应(AR)的关系。方法:以BN大鼠和Lewis大鼠分别为供受体,采用改良法建立大鼠肾移植模型。实验动物随机分为急性排斥反应组(ARx,BN Lewis)和对照组(con,Lewis Lewis),每组15只大鼠。于移植术后第3、5、7天每组分别处死5只大鼠并采用酶联免疫吸附实验(ELISA)方法检测其血清肌酐(Cr)、NGAL和CXCL10表达,对移植肾分别作病理分析。结果:根据AR严重程度半定量评分,ARx组大鼠在肾移植术后第3、5、7天分别发生轻、中、重度排斥,对照组无明显排斥现象。ELISA结果显示在肾移植术后第3、5、7天,即急性排斥早期血清Cr、NGAL和CXCL10表达较对照组显著升高(P<0.01),且呈逐渐上调趋势。结论:NGAL和CXCL10是有价值的AR标志物。ELISA法测定血清NGAL和CXCL10表达可以较敏感反映肾移植术后AR的发生,其为非侵袭联合监测方式预测或诊断肾移植术后AR发生提供了新的思路。
