摘要
目的:为研究SLA与抗病育种和经济性状的关系提供理论依据。方法:采用限制性内切酶HaeⅢ对大白猪、长白猪、杜洛克猪和马身猪SLA-DQB基因外显子2的273bp扩增产物多态性进行分析。结果:共检测到A,B,C和E等4个等位基因和AA、BB、AB、AC、AE等5种基因型。在大猪和马身猪中,A等位基因频率较高,分别为0.528和0.587;在长白猪中,检测到3个等位基因A、B和E,频率分别为0.611、0.278和0.111;而在杜洛克中,仅存在B等位基因。适合性检验表明,大白猪和杜洛克猪在该位点处于Hardy-Weinberg平衡状态(P>0.05)。结论:SLA-DQB基因外显子2多态性在4个猪种具有明显差别。
Objective: Theoretical basis were provided for the study on relationships between swine leukocyte antigen (SLA) and disease - resistant breeding and economic traits. Method: In present research, the exon 2 of SLA - DQB gene was amplified and a uniform fragment of 273bp was obtained in Large white,Landrace,Duroc and Mashen Pigs. The polymorphisms of exon 2 of SLA - DQB gene were detected by the PCR - RFLP with the restrictive enzyme Hae llI digestiorr Result: There were 4 alleles and 5 genotypes. A is the favorable allele in Large white and Mashen Pigs and the gene frequencies are 0. 528 and 0. 587 respectively; Three alleles ( A , B and E) were found in SLA- DQB locus in Landrace breeds, their frequencies are 0. 611,0. 278 and 0. 111 respectively. Only B allele was detected in Duroc population. The results of Х^2 showed that four alleles of this locus fit with Hardy - Weinberg equilibrium in Large white and Duroc Pigs. Conclusion: The obtained results suggest that this has a effective difference in the polymorphism of SLA - DQB exon 2 in 4 pig pop- ulations.
