摘要 目的探讨ghrelin对大鼠消化间期复合肌电活动(IMC)的影响及其机制。方法采用免疫组织化学和图像分析方法观察ghrelin受体(生长激素促分泌素受体,GHS-R)在大鼠消化道的分布特征;采用多导生理记录仪监测大鼠消化间期IMC,观察静脉给予ghrelin对胃肠IMC的影响;采用放射免疫法测定静脉给予ghrelin前后,IMC不同时相的血浆胃动素浓度。结果①GHS-R在胃肠道的肠肌丛和黏膜下丛有表达,胃腺体均有GHS-R免疫强阳性广泛表达,在肠腺体的阳性表达较少。②静脉给予ghrelin促进胃肠IMC,IMC周期缩短,Ⅲ相时程缩短,Ⅲ相占IMC周期百分比无显著性改变;静脉注射ghrelin后,血浆胃动素周期性波动规律不受影响,各相应时相胃动素浓度与ghrelin作用前比较无显著性差异。结论 GHS-R主要分布于大鼠消化道的肠肌丛和黏膜下丛,在胃腺体有免疫强阳性表达,在小肠腺有少量阳性表达。Ghrelin可促进胃肠运动,此作用与胃动素没有明显关系。 Objective To investigate the effects and mechanism of ghrelin on gastrointestinal interdigestive myoelectrical complex (IMC) in rats. Methods Immunohistochemistry and image analysis were applied to investigate the distribution characteristics of ghrelin receptor GHS-R in the center and the digestive tract. Rats received IMC recording by the multilead physiological recording system during fasting state before and after ghrelin injection. Radioimmunity was applied to measure the plasma motilin level before and after ghrelin administration. Results (i) GHS-R immunopositive expression appeared in the myenteric and submucous plexus of the digestive tract as well as in the gastric glands with small amounts in the intestinal glands. @ The [MC cycle length and the duration of phase I1] were decreased after ghrelin injection, without affecting the percentage of phase I]I in the IMC cycle. After intravenous injection of ghrelin, the cyclical fluctuation of motilin was not affected, and the motilin level differed little in each corresponding phase compared with that before ghrelin administration. Oonclusion GHS-R is distributed mainly in the myenteric and submucous plexus of the digestive tract. Its strong immunopositive expression is distributed mainly in gastric glands with a few weak immunoreactive cells scattered throughout intestinal glands. Ghrelin appears to be closely related to gastrointestinal motility. Its excitatory effect has no relationship with the plasma motilin level.
