反相高效液相色谱法检测原料乳中的硫氰酸钠 被引量:7

摘要 硫氰酸钠具有一定的抑菌作用,但属于对人体有毒有害的非食用添加物质。本研究开发并验证一种简单、重现性好的测定原料乳中硫氰酸钠的反相高效液相色谱法。样品采用温水溶解,超声波提取,乙酸锌和亚铁氰化钾沉淀蛋白质;测定的最佳色谱条件为:以磷酸盐缓冲液∶乙腈=95∶5(V/V)为流动相等度洗脱,流速为0.8 mL/min,色谱柱温度30℃,检测波长218 nm;硫氰酸钠的检出限为0.484 mg/kg,验证测定结果的相对标准偏差为1.55%,平均加标回收率为82.26%。该法测定样品的预处理简单,检测的精密度、回收率和灵敏度高,适用于原料乳中硫氰酸钠的定量分析。 Sodium thiocyanate had the certain effects of bacteriostasis, but it belonged to the non edible substances which was toxic and harmful to the human body. A simple and reproducible reversed-phase (RP) high-performance liquid chromatography (HPLC) method for determination of sodium thiocyanate in raw milk was developed and validated. The sample was dissolved with warm water, extracted with ultrasonic, and the protein was precipitated with zinc acetate and potassium ferrocyanide. The optimum chromatographic conditions were as follows: taking phosphate buffer-acetonitrile (95:5, V/V) as the mobile phase, the flow rate was 0.8 mL/min, temperature of chromatographic column was 30 ℃, and the detection wavelength was 218 nm. The detection limit of sodium thiocyanate was 0.484 mg/kg, the relative standard deviation of measurement results was 1.55%, and the average recovery was 82.26%. This method was suitable for the quantitative analysis of sodium thiocyanate in raw milk, the pretreatment was simple, and the precision, recovery rate and sensitivity was high.
