温和灸对动脉粥样硬化兔血清淀粉样蛋白、白介素-6、肿瘤坏死因子-α及白介素-10水平的影响 被引量:19

Effects of Mild Moxibustion Intervention on Serum and Liver Amyloid-A Protein,Serum IL-6,TNF-αand IL-10Contents in Atherosclerosis Rabbits

摘要 目的:观察温和灸对动脉粥样硬化(AS)兔血清淀粉样蛋白(SAA)、白介素(IL)-6、肿瘤坏死因子(TNF)-α、IL-10含量及肝组织淀粉样蛋白表达的影响,探讨温和灸治疗AS的抗炎机制。方法:将40只日本大耳白兔随机分为正常组、模型组、温和灸组及药物组,以高脂饲养及免疫损伤法制备AS模型。温和灸组艾灸"神阙"及"后三里",每日1次,连续14周;药物组以洛伐他丁每日10mg/kg灌胃。采用ELISA法检测血清SAA、IL-6、TNF-α、IL-10水平,观察颈总动脉组织形态学变化,Western blot法检测各组肝淀粉样蛋白表达。结果:模型组兔血清SAA、IL-6、IL-10、TNF-α明显高于正常组(P<0.01,P<0.05);温和灸组及药物组血清SAA、IL-6、TNF-α较模型组降低(P<0.05,P<0.01),IL-10较模型组升高(P<0.05,P<0.01)。模型组动脉内膜下有大量含脂质泡沫细胞增生;温和灸组及药物组泡沫细胞增生减少。模型组兔肝组织淀粉样蛋白表达明显高于正常组(P<0.05);温和灸组和药物组与模型组比较,肝组织淀粉样蛋白表达明显降低(P<0.01)。温和灸组与药物组比较,各指标的差异均无统计学意义(P>0.05)。结论:温和灸组及药物组均能通过调节相关抗炎因子和促炎因子,达到治疗AS的作用,温和灸组与药物组疗效相当。 Objective To observe the effect of mild-moxibustion on serum amyleid-A (SAA) protein, cytekine contents and liver amyleid-A protein expression in atherosclerosis rabbits, so as to explore its anti-inflammation mechanism, Methods A total of 40 Japanese male rabbits were equally randomized into normal control, model, mild moxibustion and medication (Iovas- tatin) groups. The atherosclerosis model was established by feeding the animals with high-fat diet for 14 weeks and intravenous injection of bovine serum albumin (250 mg/kg, once every week, 3 times altogether). Mild moxibustion was applied to “Shen- que” (CV 8), “Housanli” (ST 36) for 10 min, once daily for 14 weeks beginning from the first day of modeling. The SAA, serum IL-6, TNF-α and IL-10 contents were determined by double antibody sandwich method, and liver amyloid-A protein expression was detected by Western blot. The pathological changes of the cervical common artery were detected by H. E. staining. Results In comparison with the normal control group, SAA, serum IL-6, IL-10 and TNF-α contents and liver amyloid-A protein expression le- vel were significantly increased in the model group (P〈0.01, P〈0.05). Compared to the model group, SAA, serum IL-6 and TNF-α contents, and liver amyloid-A protein expression level were considerably down-regulated and serum IL-10 content was ob- viously up-regulated in the moxibustion and medication groups (P〈0.05, P〈0.01). No significant differences were found be- tween the moxibustion and medication groups in the five indexes (P〉0.05). In addition, the pathological changes of the cervical common arterial endomembrane in the moxibustion and medication group were relatively milder, suggesting a protective effect of moxibustion on liver tissue in atherosclerosis rabbits. Conclusion Mild moxibustion can relieve liver injury in atherosclerosis rabbits, which is associated with its effects in suppressing serum inflammatory cytokines and liver amyloid-A protein expression and up-regulating anti-inflammatory cytokine IL-10 level.
