摘要
目的:探讨槲皮素对非沉积高尿酸血症致肾损伤的保护作用。方法:成年雄性Wistar大鼠随机分成对照组、模型组、槲皮素组。氧嗪酸钾制备大鼠非肾脏沉积高尿酸血症肾损伤模型,槲皮素治疗5周,测定24h尿蛋白、血清、肾组织生化指标及肾脏病理学研究。结果:槲皮素组与模型组相比血尿酸、尿素氮、肌酐水平降低;模型组肾组织及血清白介素-6、肿瘤坏死因子-α水平均明显高于对照组;槲皮素组肾组织及血清白介素-6、肿瘤坏死因子-α水平均明显低于模型组;模型组肾组织及血清超氧化物歧化酶活性明显低于对照组,而丙二醛含量明显高于对照组;槲皮素组的肾组织及血清超氧化物歧化酶活性均明显高于模型组,丙二醛含量均明显低于模型组。槲皮素组的肾脏病理损伤也获得缓解。结论:槲皮素对非沉积高尿酸血症肾损伤具有保护作用,可能与槲皮素的降尿酸、抗炎、抗氧化作用有关。
Objective:To discuss the protective effect of quercetin on renal injury induced by non-crystal hyperuricemia.Methods:Male adult wistar rats were randomly assigned into three groups: control group,model group and treatment group.Daily gavage with oxonic acid at a dose of 750 mg/kg body weight was used to build non-crystal hyperuricemic rat models.Rats in the treatment group received oxonic acid as manner as model group,simultaneously they were given quercetin at a dose of 150 mg/kg body weight daily by gastric gavage.The experiment lasted for five weeks.Biochemical and pathology study were carried out.Results:Compared with control group,levels of uric acid,creatinine,blood urea nitrogen and urine protein in model group were obviously increased.Both serum and renal interleukin-6,tumor necrosis factor-α and malondialdehyde in model group were significantly higher than those in control group,associated with lower activity of superoxide dismutase.At the same time,there was an apparently pathological injury discovered in model group.Compared with model group,levels of uric acid,creatinine,blood urea nitrogen and urine protein in model group were obviously decreased.Both serum and renal interleukin-6,tumor necrosis factor-α and malondialdehyde in treatment group were significantly lower than those in model group,associated with higher activity of superoxide dismutase.Simultaneously,the pathological injury obtained relief.Conclusion:Quercetin can protective kidney from renal injury induced by non-crystal hyperuricemia.And the protective effect may be associated with ability of reducing level of uric acid,antiinflammatory and antioxidative effect.
