摘要
试验以缓慢试管苗生长为策略,建立合理使用山梨醇保存试管苗慢性生长体系,通过适宜的培养温度和合理的山梨醇浓度使用,使试管苗培养周期由原来的2~3个月延长至1年,为大规模保存马铃薯试管苗提供技术基础。试验结果表明:添加40 g/L山梨醇的培养基结合10℃培养室温度,能够使多数马铃薯试管苗保存延长至1年,从而能有效节省成本,减少由于频繁转接所引起的污染及品种混杂的发生。
The aim of this research was to establish slow growth system of plantlets in vitro by addition of sorbitol to medium based on the strategy of slow growth, and extend the subculture cycle from 2-3 months to 1 year by optimization of sorbitol concentration and culture temperature, thereby providing technique support for preservation of plantlets in vitro on large scale. The results indicated that use of medium supplemented with 40 g/L sorbitol, combined with 10℃ culture temperature, could extend the subculture cycle to 1 year for some varieties tested, therefore reducing cost, and avoiding the contamination and mixture due to frequent subculture.
