摘要
目的 研究鼻咽癌细胞bcl 2基因异常与EB病毒感染的关系。方法 采用半套式原位聚合酶链反应和免疫组织化学技术对 41例鼻咽癌组织中bcl 2基因重排、bcl 2蛋白及EB病毒潜伏膜蛋白 (LMP 1)的表达进行原位观察。结果 41例鼻咽癌中发生bcl 2基因重排 6例 ,阳性率为 14 .6 %。bcl 2蛋白表达率为 82 .9% (34/41) ,LMP 1的阳性率为 5 1.2 % (2 1/41) ,bcl 2基因异常 (重排和蛋白异常表达 )与LMP 1的阳性率无显著性差异 (P >0 .0 5 ) ,bcl 2 /JH融合基因形成与bcl 2蛋白表达无显著性差异(P >0 .0 5 )。结论 bcl 2基因在鼻咽癌发生过程中可能起一定作用。mbr和mcr区域同为bcl 2基因在鼻咽癌的 2个重要断裂区 ;bcl 2基因重排可能不是引起bcl 2蛋白异常表达的惟一原因 ;EBV感染对bcl 2基因重排和bcl 2蛋白过表达可能无影响。
Objective To study the relationship between bcl 2 gene aberrantion and EBV infection in nasopharyngeal carcinoma(NPC).Methods bcl 2/JH fusion gene,bcl 2 protein and latent membrane protein 1(LMP 1) in 41 cases of NPC were detected by semi nested in situ PCR and immunohistochemistry technique.Results 14.6%(6/41) of NPC were found to have bcl 2 gene rearrangement.34 cases of NPC expressed bcl 2 protein,the positive incidence was 82.9%.The positive rate of LMP 1 was 51.2%(21/41).There was no correlation between EBV infection and bcl 2 gene aberrant(gene rearrangement and protein overexpression).Conclusion These results suggest that bcl 2 gene might play an role in the pathogenesis of NPC.Mbr and mcrare should be the two important breakpoints of bcl 2 gene in NPC;Bcl 2 gene rearrangement may not the only cause leading to bcl 2 protein overexpression.The results also indicate that EBV infection might not influence on bcl 2 gene rearrangement and bcl 2 protein overexpression.
