摘要
目的 探讨康莱特诱导肿瘤细胞凋亡的作用机制。方法 应用形态学、流式细胞术、DNA凝胶电泳、AnnexinV标记等方法检测细胞凋亡的发生 ,应用RT PCR检测凋亡相关基因Fas、bcl 2、c myc表达的变化。 结果 康莱特对人早幼粒白血病细胞系HL6 0细胞的生长有明显的抑制作用 ,并诱导肿瘤细胞发生凋亡 ;细胞固缩 ,核染色质聚集或断裂 ;DNA凝胶电泳显示清晰的DNALadder ;流式细胞术检测在G1期之前出现sub G1峰 ,凋亡率最高为 12 .0 5 % ;AnnexinV标记的方法检测凋亡时发现坏死和凋亡共存。在HL6 0凋亡过程中 ,凋亡相关基因Fas转录水平比用药前增强 ,c myc和bcl 2含量变化不大。 结论 除了坏死 ,凋亡也是康莱特抑瘤机制之一。康莱特诱导HL6 0细胞凋亡主要与促进Fas基因表达有关。
Objective To study the anti tumor effect of kanglaite and its mechanism.Methods Morphological study,DNA gel electrophoresis,flow cytometry and fluorescein labelled annexin v were used to detect apoptosis,RT PCR to detect the apoptosis related gene.Results HL60 cells treated with kanglaite were induced to apoptosis.The morphology of HL60 cells changed as nuclear chromsomal condensation and segmentation;A typical ladder pattern fragmention were observed by DNA gel electrophoresis;Sub G 1 peak was found by flow cytometry.Fas gene was enhanced,while no difference about c myc and bcl 2 gene was found.Conclusion Apoptosis and necrosis of human tumor cells is the key mechanism for kanglaite′s killing action in the treatment of cancer.
