摘要
研究喜树碱(Cam)或和环胞菌素A(CyA)合用能否诱导抗三尖杉酯碱的人白血病HL60细胞(HR20)凋亡.采用细胞计数法、染色质凝集观察、流式细胞术和DNA琼脂糖凝胶电泳进行研究.Cam诱导敏感HL60细胞凋亡;作用HR20细胞24h,仅将细胞阻断于G2期;0.2,1mg·L-1Cam作用48h后,部分细胞发生凋亡.1mg·L-1CyA明显增加柔红霉素在HR20细胞内的积聚,但并不增加Cam诱导的凋亡细胞数量.结果表明:Cam诱导HR20细胞发生细胞凋亡需较长的时间,其作用不受CyA的影响.
The aim is to study whether camptothecin(Cam) or combining with cyclosporin A (CyA) induce apoptosis in the harringtonine-resistant human leukemia HL60 cells(HR20). The methods of nuclear chromatin condensation observation, flow cytometry and DNA agarose gel electrophoresis were used. Cam induced a typical apoptosis in sensitive HL60 cells, but arrested the HR20 cells at G2 phase at 24h. When the HR20 cells continuously treated with Cam 0.2, 1 mg ·L-1for 48 h, apoptosis occurred. CyA 1 mg·L-1 apparently increased the daunorubicin accumulation in HR20 cells, but did not increase the percentage of apoptotic cells. The results demonstrated that the induction of apoptosis by Cam in HR20 cells needed a longer time, and its action was not affectedby CyA.
