摘要
目的研究原儿茶酸(protocatechuic acid,PCA)对小鼠乳腺癌细胞(4T1)增殖和转移的影响。方法体外培养4T1细胞。实验分组:溶剂对照组(DMSO)和PCA组(浓度为1、10、100、250、500、1000nmol/L)。MTT法检测PCA对细胞增殖的影响;划痕实验和Transwell小室法观察PCA对细胞迁移和侵袭能力的影响;蛋白印迹法和荧光定量PCR法分析PCA对基质金属蛋白酶-2(MMP-2)蛋白和mRNA表达的影响。结果1~1000nmol/LPCA对4T1细胞增殖无影响;500、1000nmol/LPCA能够抑制4T1细胞的迁移和侵袭,与对照组相比,差异均具有统计学意义(P〈0.05);PCA对4T1细胞MMP-2蛋白和mRNA表达均无影响。结论PCA能够抑制小鼠乳腺癌细胞的迁移和侵袭,其作用机制可能与MMP-2无关。
Objective To investigate the anti-proliferative and anti-metastatic effects ofprotocatechuic acid (PCA) on mouse breast cancer cell line 4T1. Methods 4T1 cells were divided into seven groups: control group (DMSO), PCA groups (1, 10, 100, 250, 500, 1000 nmol/L). MTT assay was explored to evaluate the effect of PCA on proliferation of 4T1 cells. Wound healing assay and Transwell assay were applied to evaluate the inhibitory effect of PCA on the metastatic capacity of 4T1 cells. Western blot and Real-Time quantitative PCR were utilized to detect the effect of PCA on MMP-2 expression at protein and mRNA levels. Results PCA ranging from 1 to 1000 nmol/L had no significant effect on proliferation of 4T1 cells. PCA at 500 nmol/L or 1000 nmol/L significantly suppressed the migration and invasion of 4T1 cells as compared with the vehicle DMSO (P〈0.05). However, PCA at any tested concentrations had no significant effect on MMP-2 expression at both protein and mRNA levels. Conclusion PCA has anti-metastatic effect on 4T1 cells independent of MMP-2 expression.
