摘要
为了检测镍冶炼烟尘处理的NIH3T3细胞的某些生物学特征 ,采用刀豆素A (ConA)凝集试验 ,分别用两份镍冶炼烟尘与小鼠NIH3T3细胞接触 2 4小时 ,培养 2 1天后观察与ConA的凝集反应。以N -甲基 -N′ -硝基 -N -亚硝基胍 (MNNG)为阳性对照 ,二氧化钛 (TiO2 )为阴性对照。结果表明 ,用镍冶炼烟尘和MNNG处理的细胞均与ConA发生了凝集 ,呈剂量 -依赖关系。用TiO2 处理的细胞凝集反应为阴性。镍冶炼烟尘处理的细胞与ConA发生凝集意味着该细胞的生物学特征发生了改变。这一变化提示两份受检样品可能存在致癌活性 。
The objective of this study is to test the biological feature of mouse NIH3T3 cells treated with nickel refining dusts.ConA agglutinative test was applied.Mouse NIH3T3 cells were treated respectively with each of two nickel refining dust samples for 24 hours and incubated for 21 days.The agglutinative reaction of cells to ConA was observed.MNNG was selected as a positive control and TiO\-2 as a negative control.The results showed that positive reaction to ConA occurred in the cells treated with nickel refining dust or MNNG in a dose dependent manner,whereas reaction in the cells treated with TiO 2 was negative.Agglutinative reaction of ConA to cells treated with nickel refining dusts suggested an alteration of biological feature in the attacked cells.This indicates that the tested materials may have carcinogenic activity and are worth further toxicological testing and evaluation.
