羧甲基壳聚糖氟缓释膜的制备及性能检测

摘要 目的:制备多孔羧甲基壳聚糖氟缓释膜,并对其相关性能检测和体外释药考察。方法:采用冷冻干燥法制备羧甲基壳聚糖氟缓释膜,不同比例氟化钠作为有效药物成分,并用聚乙烯吡咯烷酮作为致孔剂。以孔隙率和释氟率作为指标,筛选出最优比例;扫描电镜观察缓释膜的形态;无水乙醇液体置换法检测缓释膜的孔隙率;氟离子标准曲线考察缓释膜的体外释药性。结果:羧甲基壳聚糖/聚乙烯吡咯烷酮/氟化钠比例为1∶1∶1时,制备的多孔氟缓释膜最为理想,孔隙率为(23.4±2.3)%,释氟率为0.22 mg·d-1。结论:本实验制备的多孔羧甲基壳聚糖氟缓释膜具有孔隙率适中,释氟率稳定,体外显著延缓药物释放,并期待进一步用于覆盖义齿的根面继发龋的预防研究。 OBJECTIVE To prepare porous carboxyrnethyl chitosan(CMC)/fluorine controlled-release films, and to study their performance characterization and in vitro release. METHODS Porous carboxymethyl chitosan/fluoride controllecl release film was prepared by freeze-drying process, different proportion of sodium fluoride was taken as an effective ingredient, while polyvinylpyrrolidone(PVP) as porogen. The porosity and fluorine release rate were chosen as indexes to screen the optimal for- mulation of the preparation. The morphology of membrane was observed by scan electron microscope; with anhydrous ethanol liquid displacement method to detect the porosity; with fluorine ion standard curve to study its drug release properties in vitro. RESULTS The optimal porosity and fluorine release rate were determined when the ratio of CMC to PVP to sodium fluoride was 1 : 1 : 1, the porosity was (23.4 ±2. 3)% and fluorine release rate was 0. 22 mg. d-1. CONCLUSION The porous carboxy- methyl chitosan/fluoride controlled release films had optimal porosity, and fluorine release rate was stable, and delayed drug in vitro release was valuable to be studied further for prevention of overdenture secondary root surface caries.
