摘要
目的探讨洛铂对人胃癌细胞株SGC7901放射敏感性的影响。方法 MTT法获得洛铂对人胃癌细胞株SGC7901的半数抑制浓度(IC50),分A组(洛铂4.0μg/ml+0、2、4、6、8Gy照射)和B组(0、2、4、6、8Gy照射),计算洛铂对细胞的放射增敏比(SER)。人胃癌细胞株SGC7901分为洛铂4.0μg/ml+4Gy照射(C组)、4Gy照射(D组)和空白对照(E组),流式细胞术检测三组细胞凋亡率,Western blot测定三组细胞凋亡相关蛋白B细胞淋巴瘤/白血病-2(Bcl-2)、Bcl-2相关X蛋白(Bax)和活化型半胱天冬酶-3(Caspase-3)表达水平。结果洛铂的IC50为20.66μg/ml,洛铂的SER为1.08。C、D组细胞凋亡率高于E组(P<0.01),C组凋亡率高于D组(P<0.05)。C、D组Bcl-2表达低于E组,而Bax、活化型Caspase-3表达高于E组(P<0.01)。C组Bcl-2表达低于D组,而Bax、活化型Caspase-3表达高于D组(P<0.05)。结论洛铂对人胃癌细胞株SGC7901具有放射增敏作用。其作用机制可能是抑制SGC7901细胞Bcl-2表达,激活Bax表达,与线粒体通路Bcl-2/BaxCaspase信号传导有关。
Objective To explore the effect of lobaplatin(LBP) on the radiosensitivity of human gastric carcinoma cell line SGC7901. Methods The 50% inhibitory concentration(IC50) of LBP on human gastric carcinoma cell line SGC7901 was obtained by MTT. The sensitization enhancement ratio (SER) was calculated in two groups of A(treated with LBP 4.0μg/ml and 0, 2,4, 6,8 Gy) and B (treated with 0, 2,4, 6,8 Gy). The apoptosis rate and expressions of B cell lymphoma/leukemia-2 (Bcl-2) ,Bcl-2 X-related protein(Bax) and cleaved Caspase-3 were detected by flow cytometry and Western blot, respectively, in three groups of C (treated with LBP 4. 0 μg/ml and 4 Gy), D (treated with 4 Gy alone) and E(blank control). Results The ICs0 and SER of LBP were 20. 66μg/ml and 1.08. The apoptosis rate in groups of C and D was higher than that in group E(P〈0. 01), which in group C was higher than that in group D(P〈0. 05). Compared with group D, the expression of Bcl-2 was lower, while expressions of Bax and cleaved Caspase-3 were higher in group C (P〈0. 05). Compared with group E, the expression of Bcl-2 was lower,while the expressions of Bax and cleaved Caspase-3 were higher in groups of C and D (P〈0. 01 ). Conclusion LBP can enhance the radiosensitivity of human gastric cancer cell line SGC7901, which may be related to downregulating P, cb2 expression and upregulating Bax expression via Bcl-2/Bax-Caspase signal pathway.
