VEGF在大鼠脊髓损伤后股骨干骨痂形成过程中的表达 被引量:4

VEGF Expression in Femur Callus Formation after Spinal Cord Injury with Femoral Shaft Fracture in the Rat

摘要 目的 探讨大鼠在脊髓损伤伴骨折中骨折愈合速度增快的原因.方法 来自甘肃省中医学院动物实验中心的12周龄大鼠40只,使用抽签法分为两组,实验组为脊髓损伤伴股骨干骨折20只,对照组为单纯股骨干骨折20只,制作股骨干骨折与脊髓损伤的模型.术后分别在1周、2周、3周、4周给予X线摄影,及每周处死实验组和对照组大鼠各5只,截取骨痂,用免疫组织化学染色方法检测血管内皮生长因子(VEGF)表达.结果 X线摄影显示,实验组骨痂形成较对照组骨痂的形成在同一时间点早且数量增多.免疫组织化学结果显示,实验组术后1 ~4周骨痂中VEGF阳性细胞百分率分别高于同一时间点对照组(P<0.05).结论脊髓损伤对骨折愈合有明显的促进作用,这与脊髓损伤后VEGF表达水平升高有关. Objective To explore reasons of accelerating fracture healing after spinal cord injury with femoral fractures in rats. Methods Forty rats aged 12 weeks from Animal Experiment Center of Gansu Tra- ditional Chinese Medicine College were randomly divided into 2 groups by ballot method, experimental group of 20 rats with spinal cord injury with femoral fracture, control group of 20 rats with femoral shaft fractures alone. X-ray for the rat models was taken at 1 week,2 weeks,3 weeks ,4 weeks after surgery respectively, and 5 rats of experimental group and control group each were killed every week to intercept bony callus, and detect expression of VEGF in the bony callus with immunohistochemistry. Results X-ray showed that callus formation of the experimental group was early and increased than control group at the same time point. Immu- nohistochemical results showed that VEGF expression was significantly higher in the bony callus of experi- mental group than the control group at the same time point from 1 week to 4 weeks after surgery( P 〈 0.05 ). Conclusion The spinal cord injury can significantly stimulate fracture healing, which is associated with elevated VEGF expression levels.
