摘要 生物活性导向分离海洋微生物次级代谢产物是海洋天然产物研究的热点,对海洋链格孢属真菌Alternariasp.MNP801菌体甲醇提取物建立体外抗氧化和抗肿瘤双模型活性测试,从中导向分离得到3个化合物:5α,8α-表二氧麦角甾-6,22-二烯-3β醇(Ⅰ)、氧杂蒽酮(Ⅱ)、硬脂酸(Ⅲ).其中化合物Ⅰ为首次从海洋链格孢属真菌中分离得到的化合物,验证实验表明化合物Ⅰ对肿瘤细胞H460,3T3,PC12,U937具有强的抑制活性,其IC50值分别为51.26,41.26,8.7和14.6μg/mL,对H460,3T3肿瘤细胞的抑制作用与对照组VP16的抑制活性相当,而对PC12,U937肿瘤细胞的抑制作用远高于对照组VP16的抑制活性,表明化合物Ⅰ具有潜在的药物开发价值. Bioactivity guide isolation of secondary metabolites from marine microorganism is a hotspot of marine natural product research.The methanol extracts of mycelium from marine fungus Alternaria sp.MNP801 have been tested by antioxidative (DPPH) and antitumor (MTT) active methods and three compounds were isolated:5α,8α-epidioxy-ergosta-6,22-diene-3β-ol (Ⅰ),xanthone (Ⅱ),stearic acid (Ⅲ).The compound I was isolated from marine fungus Alternaria sp.for the first time.The confirmatory experiment showed that compound Ⅰ had strong cytotoxicity against H460,3T3,PC12 and U937 tumor cells,its IC50 value is 51.26,41.26,8.7 and 14.6 μg/mL,respectively.The activities of compound Ⅰ against H460,3T3 tumor cell are the same as the contrast VP16.The activities of compound Ⅰ against PC12 and U937 tumor cells are higher than that of the contrast VP16.It shows that compound Ⅰ is a compound with potential medicinal applications.
