摘要
目的探讨抑制人CD74基因后影响肾癌786-O细胞侵袭能力的作用机制。方法通过慢病毒lentivirus-CD74转染肾癌786-O细胞后,MTT法观察细胞活力变化,Transwell实验观察细胞侵袭能力的变化,应用Westernblot技术检测CD74、CXCR4蛋白的变化情况。结果抑制CD74蛋白表达水平后肾癌786-O细胞活力降低,侵袭能力降低,同时伴有CXCR4蛋白的表达降低。结论通过慢病毒转染抑制CD74的表达可以抑制肾癌细胞的侵袭能力,CD74基因可能是通过调控CXCR4蛋白的变化影响细胞侵袭能力的。
Objective To study Methods Lentivirus-CD74 was used to the effect and mechanism of CD74 in inhibiting 786-0 cell invasion in renal cancer transfect renal cancer 786-0 cells,cell vitality was observed with MTT assay,cell inva- sion was observed with transwell assay, and the expressions of CD74 and CXCR4 were detected with Westernblot. Results The transfected lentivirus-CD74 786-0 ceils induced decreased ceil proliferation and invasion,and at the same time, the expres- sion of CXCR4 protein reduced. Oonclusions Transfecting CD74 with lentivirus can inhibit the cell invasion of renal cancer, indicating that CD74 may affect the cell invasion by regulating CXCR4 expression.
