摘要
目的:观察羌活地黄汤对氟氏完全佐剂诱导的类风湿性关节炎(AA)大鼠血清肿瘤坏死因子-α(TNF-α)、白介素-1β(IL-1β)、白介素-6(IL-6)的影响,探讨羌活地黄汤治疗类风湿性关节炎的可能机制,为羌活地黄汤在临床上的应用提供实验依据。方法:建立大鼠佐剂性类风湿性关节炎动物模型,将SD大鼠随机分为正常对照组、模型组、羌活地黄汤及甲氨喋呤组。采用灌胃给药方式进行治疗。采用酶联免疫吸附法检测AA大鼠血清中TNF-α、IL-1β、IL-6的水平。结果:佐剂性关节炎大鼠血清TNF-α、IL-1β、IL-6水平均高于正常对照组(P<0.01);经羌活地黄汤及甲氨喋呤治疗后,羌活地黄汤组及甲氨喋呤组的TNF-α、IL-1β、IL-6水平低于模型组(P<0.01),羌活地黄汤组与甲氨喋呤组血清TNF-α、IL-1β、IL-6水平比较差异无统计学意义(P>0.05)。结论:羌活地黄汤具有抑制AA大鼠TNF-α、IL-1β、IL-6的异常分泌的作用,可以阻止AA大鼠免疫性炎症的发展。提示羌活地黄汤对AA大鼠TNF-α、IL-1β、IL-6的影响是其治疗RA的可能机制。
Objective：To observe the effect of Qianghuo Dihuang Decoction on the serum levels of tumor necrosis factor al- pha （TNF-α）, interleukin-1β（IL-1β） and interleukin-6 （IL-6） in rats with Freund＇s Complete adjuvant-induced rheumatoid arthritis, and to explore the possible mechanism of Qianghuo Dihuang Decoction in the treatment of rheumatoid arthritis, so as to provide experimental basis for the application of Qianghuo Dihuang Decoction in clinic. Methods：The adjuvant ar- thritis rat model was established. SD rats were randomly divided into normal control group, model group, Qianghuo Di- huang Decoction group and methotrexate group. Intragastric administration method was adopted in the treatment. Enzyme- linked immunosorbent assay was used for detection of the serum levels of TNF-α, IL-1β and IL-6 in AA rats. Results.. The serum levels of TNF- α, IL- 1β and IL- 6 in model group were higher than those in normal control group （P〈0.01）. After treatment, the serum levels of TNF-α, IL- 1 β and IL- 6 in Qianghuo Dihuang Decoction group and methotrexate group were lower than those in the model group （P〈0.01）. There was no significant difference between Qianghuo Dihuang De- coction group and the methotrexate group in the serum levels of TNF- α, IL- 1 β and IL- 6 （P 〉 0.05）. Conclusion：Qiang- huo Dihuang Decoction can inhibit the abnormal secretion of. TNF- α, IL- 1 β and IL- 6 in AA rats, so that it can prevent the development of immune inflammation in AA rats. Our experimental results showed that the effect of Qianghuo Dihuang Decoction on serum levels of TNF- α, IL- 1β and IL- 6 is the possible mechanism in the treatment of RA.
