摘要
[目的]用基因芯片技术研究高低转移人卵巢癌细胞系(HO-8910PM和HO-8910)基因表达谱差异,筛选与转移相关的 基因。[方法]分别抽提高低转移人卵巢癌细胞和对照正常卵巢上皮的总RNA并纯化mRNA;分别将等量的mRNA逆转录合成以 Cy5和Cy3标记的cDNA做探针,分别混合后在2张含有4096条双点人类全长基因的芯片上进行杂交。扫描仪扫描芯片荧光信号 图像,用软件对扫描图像进行数字化处理和分析。[结果] HO-8910细胞与正常卵巢上皮比较差异3倍以上共有355个基因; HO-8910PM细胞与正常卵巢上皮比较差异3倍以上共有323个基因。HG-8910PM与母系HO- 8910比较差异2倍以上共有163个 基因,差异3倍以上共有21个基因。[结论]两株人卵巢癌细胞与正常卵巢上皮细胞基因表达谱存在差异,提示这些基因与卵巢 癌的发生和发展有关;HO-8910PM与 HO-8910比较存在差异的基因可能与高转移特性相关。
Purpose] TO study the gene expression difference of high (HO-8910PM) and low (HO-8910) metastatic human ovarian carcinoma cell line and to screen novel associated genes by cDNA microarray. [Methods] The cDNA retro-transcribed from equal quantity mRNA derived from high and low metastatic tumor cells or normal ovarian tissues, which were labeled with Cy5 and Cy3 fluorescence as probes .The mixed probes were hybridized with two pieces BioDoor 4096 double dot human whole gene chip. It was scanned by laser scanner. The acquired image was analyzed by software. [Results] By applying the cDNA microarray we found: A total of 355 genes whose expression level were larger than 3 times in HO-8910 cell and normal ovarian epithelium cell was screened out. A total of 323 genes whose expression level were larger than 3 times in HO-8910PM cell and normal ovarian epithelium cell was screened out. A total of 163 genes whose expression level were larger than two times in HO-8910PM cell and its mother line HO-8910 cell was screened our, and 21 genes whose expression level were larger than three times. [Conclusion] cDNA microarray technique is effective in screening the differential gene expression between two human ovarian cancer cell line (HO-8910PM; HO-8910) and normal ovarian epithelium cell. It suggests that these genes related to genesis and development of ovarian carcinoma.One hundred and sixty three genes whose expression level are larger than 2 times in HO-8910PM and HO--8910 cell line are found. It indicates these genes probably related to the potency of high metastasis.
