农杆菌介导的桑树遗传转化条件的研究 被引量:5

摘要 本实验探讨了多种因子对桑树遗传转化的影响,结果表明:菌株LBA4404 比C58Cl转化效率高;不同外植体转化效率有差异,以叶盘、茎尖和愈伤组织为受体转化频率分别为8.7% 、5.6% 和2.8% ;农杆菌菌液浓度、感染时间、共培养时间等对桑树叶盘转化效率有一定影响;AgNO3 对遗传转化具有明显的促进作用。 In this paper,several factors which affecting genetic transformation of mulberry mediated by agrobacterium tumefaciens were studied.It showed that the strain of LBA4404 was more efficient than that of C58Cl.The transformation frequency were different among the explants,such as leaves discs,shoots and calli,it 8.7%,5.6% and 2.8% respectively.The consistence of agrobacteria,the time of dipping the leaves discs of the mulberry in the agrobacteria and the co cultivation time had some effects on the transformation frequency.In addition,AgNO 3 had prominence stimulation to the transformation of mulberry.
