摘要
目的 研究 CD44粘附分子在脑膜瘤、多形性胶质母细胞瘤和脑转移瘤中的表达及其在瘤细胞转移、侵袭中的作用 .方法 采用免疫组化 SABC法检测脑膜瘤、多形性胶质母细胞瘤和脑转移瘤各 2 0例及 10例正常脑组织中 CD44 s,CD44 v3,CD44 v6和 CD44 v3- 10的表达 .结果 1CD44 s在脑膜瘤、多形性胶质母细胞瘤和脑转移瘤中的表达率分别为15 % ,10 0 %和 90 % ,其表达在前两者间有显著性差异 (P<0 .0 1) ;2 CD44 v3,CD44 v6和 CD44 v3- 10在脑膜瘤和多形性胶质母细胞瘤中无阳性表达 ,在脑转移瘤中表达率分别为40 % ,5 0 %和 75 % (P<0 .0 1) ;3CD44 s,CD44 v3,CD44 v6和CD44 v3- 10在正常脑组织中均无阳性表达 .结论 1CD44 s蛋白可能在多形性胶质母细胞瘤的脑内侵袭过程中起重要作用 ;2 CD44 v蛋白可能在部分脑转移瘤向颅内转移的过程中起作用 。
AIM To investigate the expression of CD44 in meningiomas,glioblastomas and brain metastases,and its role in metastasis or invasion of these tumors. METHODS Expression of CD44 standard form (CD44s), the variant isoforms containing the domain encoded by variant exon 3 (CD44v3), variant exon 6 (CD44v6) and variant exon 3 to 10 (CD44v3 10) were detected by techniques of immunohistochemistry in paraffin embedded tissues from 20 meningiomas, 20 glioblastomas multiforme(GMs), 20 brain metastases and 10 normal brains. RESULTS CD44s was expressed in 15% of meningiomas, 100% of GMs and 90% of brain metastases, and there was a significant difference between the expression of CD44s in meningiomas and that in GMs( P <0 01). The expressions of CD44v3, CD44v6 and CD44v3 10 in brain metastases were 40%, 50% and 75% respectively. None of 20 meningionas and 20 GMs expressed CD44v. There were marked differences between the expressions of CD44v in GMs and those in brain metastases ( P <0 01). None of the 10 normal brain tissues expressed CD44s, CD44v3, CD44v6 and CD44v3 10. CONCLUSION CD44s protein may play an important role in the intracerebral invasive process of GM. CD44v protein may play a role in the intracerebral spread of some of the brain metastases.Therefore,CD44v expression Should be further considered as a potential marker for diagnosis and treatment of patients with brain metastases.
