三维及二维培养的病理性瘢痕成纤维细胞内胶原合成的定量分析

摘要 目的 定量分析比较三维及二维培养的病理性瘢痕成纤维细胞内合成的前胶原蛋白。方法 以成纤维细胞为实验对象 ,应用定量免疫细胞化学技术及粘附式激光扫描细胞仪 ,检测三维及二维培养系统中成纤维细胞内的Ⅰ型前胶原蛋白。结果 三维系统中瘢痕成纤维细胞内Ⅰ型前胶原蛋白的量远低于后二维培养系统。 Objective To compare the procollagen synthesis of patho logical scar derived fibroblasts from three and two dimensional culture system .Methods Immunocytochemistry and ACAS were used to estimate procollagen in fibroblasts from three and two dimensional culture separately. Results Observations show that the procollagen synthesis in three dimensional culture system is much lower than the latter.Conclus ion The biologic characteristics of scar derived fibroblasts from th ree dimensional culture is more similar to that in vivo.
