摘要
利用SRAP分子标记技术,对苦瓜品种佳美及其亲本的DNA指纹进行分析研究,经多次试验,从108对引物中筛选出2对特征引物能同时扩增出F1代及其父母本的多态性条带,扩增产物具有高度的稳定性和重复性,构建了相应的DNA指纹图谱。对100株佳美苦瓜进行了纯度鉴定,结果有3株植株只有母本特异带而没有父本特异带,与大田检测的结果完全一致,杂交率达97%。
Using SRAP molecular marker technology,the DNA fingerprints of Jiamei bitter gourd and its parents were analyzed.After repeated experiments,2 pairs of primers were screened out from 108 primer pairs,which could amplify the characteristic polymorphic bands of F1 generation and its parents,with the PCR products possessing high stability and repeatability,and the corresponding DNA fingerprints were built.100 individuals of Jiamei bitter gourd were identified. The results showed that only 3 individuals had female-specific band without male-specific band,which agreed with the result of the field test,and the hybrid rate was 97%.
