摘要 利用果糖在碱性条件下可与Cu2+络合后在紫外区有特定吸收波长的原理,建立Cu2+络合物紫外发测定果糖含量的方法,并采用正交实验设计测定果糖含量的最佳条件。结果经正交试验选择果糖含量测定最佳试剂条件为:0.10%的硫酸铜溶液0.8 mL,0.20%的氢氧化钠溶液2.0 mL。测定波长为237 nm,果糖在0.015 mg/mL^0.050 mg/mL浓度范围内线性关系为A=17.614C-0.095 8,r=0.999 9,平均回收率为99.92%(n=9),RSD为0.58%。 To establish and validate a Ultraviolet (UV) method for determination of fructose. The content of fructose was measured by UV spectrophotometry with orthogonal test carried out because fructose reacted with Cu2+ in alkaline conditions to form a complex compound which had a specific absorption in the UV spectrophotometer. The results showed the optimum condition for determination of fructose was 0.8 mL 0.10 % CuSO4 solution with 2.0 mL 0.20 % NaOH solution. The wavelength of the complex compound was 237 nm. It showed a good linear relationship in the concentration range of 0.015 mg/mL-0.050 mg/mL (A =17.614C-0.095 8, r=0.999 9). The average recovery (n=9) was 99.92 %, and RSD was 0.58 %.
