剔毒护肝方药物血清对大鼠肝星状细胞产生Ⅰ型胶原及转化生长因子β_1的影响 被引量:7

摘要 目的:根据剔毒护肝方抗肝纤维化的主要作用机理。方法:分离正常大鼠肝星状细胞、并传代培养。用剔毒护肝方给正常大鼠灌胃,制备药物血清,温育培养细胞。用^3H-TdR掺入法和MTT比色法观察细胞增殖,ELISA法测定上清液中Ⅰ型胶原含量,貂肺上皮细胞生长抑帛MTTI地检测培养上清液中的转化生长因子β1活性。结果:剔毒护肝方药物血清能显著4抑制肝星状细胞增殖,并能抑制肝星状细胞生成Ⅰ型胶原及产生转化生长因子β1。结论:剔毒护肝方能抑制肝星状细胞增殖和分泌转化生长因子β1,养活胶原生成,从而减弱肝星状细胞的自分泌放大效应。这可能是剔毒护肝方抗肝纤维化作用的主要机制之一。 To investigate the main serapharcological mechanism of antifibrotic action of Tidu Hugan Recipe at cellular and molecular level. Methods: Hepetic stellate cells (HSC) were isolated from the nomal rats and subcultured. Nomal rats were medicated with TDHGR and their blood serum collected to prepare the medicated serum, which was incubated with the subcultured HSC. Then, observed the proliferation of the HSC by H-TdR incorporation and MTT col-orimetric assay, tested the content of collagen I in the supernatant by using the ELISA method, measured the activity of TGFpi IK the supernatant by mink lung epithelial cell (Mvliu) proliferation in hibition MTT assay. Results: TDHGR medicated serum could obviously inhibit the proliferation of HSC, the production of collagen I and the yield of TGFβ1 Conclusions: TDHGR could inhibit the proliferation of HSC and the production of collagen I , and reduce the yield of TGFP) in the HSC. The inhibition of the amplified autocrine effect of HSC may be one of the main mechanisms of TDHGR anti-hepatic fibrosis actions.
