摘要
目的 探讨颅内神经上皮源性肿瘤端粒酶活性的表达。方法 采用改良的端粒重复序列扩增法(TRAP)对新鲜冻存的 78例颅内常见肿瘤和 5例正常人脑组织标本 ,进行了端粒酶活性检测。结果 颅内常见恶性肿瘤端粒酶活性的表达率为 79% (4 2 /5 3) ,良性肿瘤的表达率则为 8% (2 /2 5 ) ,两者之间有显著差异 (P <0 .0 5 )。正常人脑组织标本均为阴性。星形细胞瘤的端粒酶表达随肿瘤恶性程度的增加而增高 ,I~II级 (6 5 % )和III~IV级 (10 0 % )的星形细胞瘤端粒酶表达阳性率之间有统计学意义 (P <0 .0 5 )。结论 端粒酶在颅内常见良、恶性肿瘤中均有表达 ,但阳性率不同 ,与肿瘤的组织学分级和生物学行为有关。
Objective To explore telomerase activity in a broad of intracranial neuroepithelial tumors. Methods 78 frozen samples of different tumors and 5 cases of normal brain tissues were analysed by a modified telomeric repeat ampli\|fication protocal(TRAP). Results Telomerase activity was detected in 79%(42/53) of malignant neoplasms, while in 8%(2/25) of benign neoplasms. There was a significant difference between 2 types of tumors(\%P<0.05\%). Telomerase activity was negative in 5 normal brain tissues. In the case of astrocytic tumors, the frequency of telomerase activity increases with malignancy. The detection rates were significantly different between grade I~II(65%) and grade III~IV(100%) of astrocytomas. Conclusion Telomerase had different expression in benign and malignant intracranial tumors, telomerase activity was correlate with histological grade and biological behaviour of tumors. [
