摘要
利用PCR方法对采自天津地区某猪场的疑似猪圆环病毒2型(PCV-2)感染的病料进行检测,并确定病料中PCV-2的分子特征。将阳性病料接种无PCV污染的PK-15细胞并传代8次,分离出l株PCV-2毒株,命名为PCV-2TJ-1,同时对其全基因组进行克隆和序列分析。结果表明,该分离株基因组全长1767bp,与我国上海LN-19株和韩国的A4099株的核苷酸同源性最高,均达到99.6%。PCV-2全基因组序列的进化分析表明,分离毒株为PCV-2b基因亚型;对分离毒株与5个不同基因亚型的10个参考株的ORF2和ORF3氨基酸序列进行比较,同源性分别为87.1%~97.4%和93.3%~99.0%。结果表明,在天津地区流行的PCV-2毒株为PCV-2b,为PCV-2的分子流行病学、遗传变异及防控奠定了基础。
Samples suspiciously infected with porcine circovirus type 2 (PCV-2) from Tianjin were detected by using polymerase chain reaction (PCR). PCV-2-positive samples were inoculated into non-PCV con- tained PK-15 cells and passaged eight times. One strain was isolated, and named PCV-2 TJ-1. The com- plete genome of PCV-2 TJ-1 strain was cloned, sequenced and analyzed. Results showed that the full length of genomic sequence of PCV-2 TJ-1 strain was 1 767 nt and the isolate shared the highest identity (99.6%) with that of China LN-19 strain and Korea A4099 strain. Phylogenetic analyses demonstrated that the isolate was classified into PCV-2b genotype. The ORF2- and ORF3-encoded amino acid sequence comparison of PCV-2 TJ-1 and 5 different genotypes of 10 PCV-2 strains showed that the homologies were 87. 1%-97.4% and 93.3%-99.0%, respectively. In conclusion, PCV-2b group strain was predominant prevailing strain in Tianjin, which may provide some basis for studying the molecular epidemiology, varia- tion and prevention of PCV-2.
