摘要
探讨适合家蝇3龄幼虫的石蜡切片制作方法,并通过HE染色和PAS染色观察家蝇3龄幼虫的组织结构及其多糖和糖蛋白的分布。实验结果显示,Carnoy固定液或加入TritonX-100的多聚甲醛固定液都适合家蝇3龄幼虫的固定,固定后切除部分表皮是保证脱水和包埋成功的关键。实验成功获得家蝇3龄幼虫石蜡切片,清楚地显示了家蝇3龄幼虫消化系统、中枢神经系统、排泄系统、血细胞、呼吸系统、表皮、肌肉和脂肪体的组织结构,在骨骼肌纤维、表皮细胞和脂肪体细胞的胞质中均见糖原颗粒,在中肠和马氏管上皮腔面有纹状缘分布。研究结果为蝇类的分类和防治提供了较系统的形态学资料,并为进一步通过原位杂交和免疫组织化学技术定位功能分子提供了可靠的实验基础。
To establish a proper method of making the paraffin section of the third instar larva of Musca domestica, and to observe the tissue structure of the third instar larva of Musca domestica and the distribution of polysaccharides and glucoprotein by hematoxylin - eosin staining and periodic acid - schiff staining. The results showed that Carnoy fixation fluid or 4% paraformaldehyde added 0.25% TritonX - 100 is suitable to fix the third instar larva of Musca domestica. The key point of dehydration and embedding was to remove parts of cuticle. The paraffin section of the third instar larva of Musca domestica was successfully made. The tissue structure of digestive system, central nervous system, excretory system, blood cell, respiratory apparatus, cuticle, muscle and epiploon was displayed clearly. The glycogen granules were found in the cytoplasm of muscle fiber, cuticle cells and epiploon cells. The striated border was observed on the epithelial cavosurface of midgut and Malpighian tubules. The research systematically provided the morphology information for classify, prevention and cure of the flies. It will benefit the molecule location in situ hybridization and immunohistochemistry technology.
