摘要
采用ISSR分子标记对福建漳平五一林场28个马尾松优良家系最优单株进行遗传分析,计算两两之间的遗传距离,进行遗传结构分析,结果表明:10条多态性引物共检测到99个位点(从250~2 300 bp),其中93个多态位点,多态位点百分率(P)为93.94%;28个马尾松无性系两两之间的分子遗传距离在0.212 1~0.565 6之间,遗传距离大于0.5的无性系对有11组,遗传距离小于0.25的无性系对有6组;在聚类分析自检举值为48处,可将28份马尾松无性系分为9类,其中7个无性系单独聚类,其他21个无性系聚成2类;遗传距离较大的无性系组分别出现在不同的聚类中,表明这些无性系组两两之间具有相对较远的亲缘关系。本研究为马尾松高世代育种群体的选育提供分子水平上的依据,为分子标记指导马尾松育种奠定了一定的研究基础。
ISSR (Intro-simple Sequence Repeat) were employed to study the genetic analysis and GD ( genetic distance) among 28 elite clones in Pinus massoniana Lamb. The results indicated that 10 polymorphism markers totally produced 99 alleles (size ranged from 250 to 2300bp) and 93 were polymorphic, the percentage of polymorphic alleles (P) was 93.94%. The GD among 28 clones ranged from 0.2121 to 0.5656. There were 11 pairs of clones' GD larger than 0.5 and 6 pairs of clones' GD less than 0.25; The 28 clones were clustered into nine groups at the value of 48, of which seven clones were clustered separately and the other 21 clones were clustered into two groups; Those pairs of clones with larger GD appeared in different groups which showed a distant relationship with each other. These findings provided some insight into the selection of elite clones in breeding population on advanced generation orchard and it would guide the breeding work ofPinus massoniana Lamb more efficiently at the molecular level.
