摘要
目的:探讨复方黄芪汤和小柴胡汤抗HCV的作用机制。方法:用不同剂量生药和不同采集时间的药物血清,处理PBK-HCV重组体转染的SMMC_7721肝癌细胞,然后用RT-PCR半定量法测定细胞内重组体mRNA瞬时表达情况。结果:复方黄芪汤对SMMC_7221细胞生长的半数抑制剂量为55mg/ml,小柴胡汤为75mg/ml,两方均不能抑制重组体mRNA表达。结论:复方黄芪汤和小柴胡汤的抗HCV作用不是通过抑制mRNA的转录,而可能是通过调节机体免疫功能发挥作用。
Aim; In order to study the anti HCV mechanism of Fufang Huangqi Decoction and Xiao Chaihu Decoction. Methods: The SMMC7721 cell line were transfected with the recombinant PBK-HCV (contained HCV cDNA 317-2009 fragment). The cell line were grown in RPMI 1640 medium with different dose decoction and different serum-phar-macon of time, and observed expressing of the recombinant mRNA by RT-PCR semi-quantityassay. Results: ID50 was 55mg/ml and 75mg/ml respectively. Two decotions could not reduced the recombinant mRNA expressing. Conclusion: Fufang Huangqi Decoction and Xiao Chaihu Decoction exert potent anti-HCV activity maybe enhances immune function of the host.
